Title: Vergleich der Transfektionseffizienz und Anzahl von generierten stabilen Klonen durch Elektroporation zirkulärer und linearer DNA in eukaryotischen Zellen

Abstract: 
Ziel dieser Arbeit ist es, eine Aussage über die Transfektionseffizienz und die Anzahl von generierten stabilen Klonen durch Elektroporation mit zirkulärer und linearer DNA in CHO-Zellen zu treffen. Die Vermutung ist, dass die Effizienz bei der transienten Transfektion mit zirkulären Vektoren höher ist als bei der Transfektion mit linearen Vektoren. Dabei sollen CHO-K1-Zellen mit einem für GFP codierenden Gen transfiziert werden. Dazu werden zwei verschiedengroße Vektoren ausgewählt, ein kleiner Vektor mit 4731 bp (pEGFP-C1) und ein großer Vektor mit 8180 bp (pCMV-GFPlong). Als Transfektionsmethode wird die Elektroporation mit dem MaxCyte®-STX™-System gewählt.
Aus den transfizierten Zellen sollen zusätzlich stabil-transfizierte Klone isoliert werden. Hier ist die Vermutung, dass die stabile Transfektion eine höhere Effizienz bei der Verwendung von linearen Vektoren hat als bei der Verwendung zirkulärer Vektoren [Oliveira et al., 2005].
Zur Verifizierung der Expression von GFP soll mit Hilfe der Fluoreszenzmikroskopie das rekombinante GFP nachgewiesen werden. Die Zellkernfärbung mit DAPI soll als Gegenfärbung eingesetzt werden.
Es soll ermittelt werden, welchen Einfluss die Konformation und die Größe des zu transfizierenden Vektors auf die Effizienz der Transfektion hat. Eine weitere Aussage ist zu treffen, welchen Einfluss die verschiedenen Vektoren und deren Konformation auf die Viabilität der transient-transfizierten Zellen hat.

In this thesis, Chinese hamster ovary cells (CHO) were transfected with the gene for the expression of GFP. Two vectors of different sizes, a small one with 4731 bp (pEGFP-C1) and a large vector of 8180 bp (pCMV-GFPlong) were used. The electroporation with the MaxCyte®-STX™-system was selected as transfection-method. GFP-expression was verified by fluorescence microscopy. The influence of the conformation and size of the vectors on the efficiency of the transient transfection as well as on the generation of stable clones was investigated. For transient transfections the viability of the cells as well as the efficiency was examined. For stable clones it was focused on the overall amount of clones and also on the number of positive clones and the homogeneity of GFP expression. Results have shown, that the efficiency of the transient transfection was about 85 %, which is slightly below the value the company MaxCyte® claims in their product discription. The viability of the transfected cells was higher than the expected value of 90 %. Furthermore the data reveals, that the size of the vectors had a higher influence on the transfection-efficiency than the conformation of the vectors. The investigation of the generated stable clones showed, that the transfection of both vectors in circular conformation resulted in 25 % homogenous clones. The linear conformation resulted in of 18 % homogenous clones. These data indicates that the conformation of the vectors has a higher influence on generating of stabile and homogenously expressing cells than the size of the vectors.
