Title: In vitro Untersuchungen zur Wirkung von nanostrukturiertem Carbon Black an Lungenepithelzellen

Abstract: 
Carbon Black ist ein nanostrukturiertes Material, das durch unvollständige Verbrennung entsteht und heute zu den meistproduzierten Industriechemikalien gehört. Sein Anwendungsspektrum ist äußerst breit und umfasst vor allem die Industrie für Gummiformteile und andere Gummiwaren. Carbon Black ist aber nicht nur in Reifen, sondern auch in unzähligen anderen Produkten wie Lacken, Farben, Hygieneartikeln, Medizin und Kosmetik zu finden. Durch zahlreiche Untersuchungen auf eine mögliche toxikologische Wirkung wurde Carbon Black von der Weltgesundheitsorganisation als potentiell kanzerogen eingestuft. Ihre geringe Größe von wenigen Nanometern lässt die Partikel in die Lunge und damit auch in die tiefen Alveolen gelangen. Da durch Umweltbelastungen viele Menschen unter Lungenerkrankungen wie Asthma leiden, soll dies durch eine Vorschädigung mit Stickstoffdioxid (NO2) und Lipopolysaccharid (LPS) nachgestellt werden.
In dieser Arbeit wurden vier Industrieruße mit unterschiedlichen Oberflächenbeschaffenheiten auf ihr toxikologisches Potential untersucht. Bei drei der vier Partikel handelte es sich um Acetylenruß, Printex®90 und Modifikationen dieses Rußes. Die modifizierten Printex®90 Ruße haben auf ihrer Oberfläche gebundene polyzyklische Kohlenwasserstoffe (PAK), Benzo(a)pyren und Nitroanthracen. Um ihr Potential auf die Lunge zu untersuchen, griff man auf die Alveolarepithelzellen A549 zurück. Zur Untersuchung der Wirkung der nanostrukturierten Carbon Black wurden zwei Testmethoden eingesetzt. Um die mögliche Toxizität auf die Zelle zu messen, wurde ein Viabilitätstest gemacht und die ausgeschüttete Interleukin 8 Konzentration der Zellen untersucht. Beim Viabilitätstest sowie bei der Auswertung des ausgeschütteten Interleukin 8 wies keins der verwendeten Ruße große Effekte bei den Lungenepithelzellen hervor.
Anhand der aufgezeichneten Ergebnisse lässt sich sagen, dass bei den verwendeten Konzentrationen der Ruße kaum eine toxikologische Wirkung vorliegt. Der geringe Effekt von Printex®90 ist auf die spezifische Oberfläche zurückzuführen.

Carbon Black is a nanostructured material, which arises through incomplete combustion and ranks among the industrial chemicals with the highest production rates.
It has a quite wide range of applications and comprises mainly the rubber industry. Carbon Black is not only found in tires, but also in countless other products such as varnishes, paints, hygiene items, medicine and cosmetics. Numerous studies on the possible toxicological effects of Carbon black lead to the classification potentially carcinogenic by the World Health Organization.
Due to their small size of a few nanometers the particles can reach the lungs and thus also the deep alveoli. Through environmental pollution many people have to deal with lung diseases such as asthma, which should be reconstructed through a pre-damage with nitrogen dioxide (NO2) and lipopolysaccharide (LPS).
In this work, four industrial carbon black particles with different surface conditions were examined on their toxicological potential. Three of the four particles were acetylene soot, Printex®90 and modifications of this carbon black. The modified carbon black particles have polycyclic aromatic hydrocarbons (PAH) Benzo(a)pyrene and nitroanthracene bound to their surface.
In order to investigate their potential in the lungs the alveolar epithelial cells A549 were used. To investigate the effect of the nanostructured carbon black two test methods have been used. To measure the potential toxicity to the cell, a viability assay was taken and distributed interleukin 8 concentration of the cells was examined. The viability assay and the evaluation of distributed interleukin 8 revealed that none of the carbon black particles showed large effects in lung epithelial cells.
Based on the recorded results it can be said that toxicological effects at the used concentrations of carbon black particles hardly exist. The small effect of Printex®90 is due to the specific surface area.
