Title: Photolytische Behandlung von Levetiracetam und Pregabalin: Abbaubarkeit und Kinetik der Muttersubstanzen und deren Phototransformationsprodukte

Abstract: 
Arzneimittelrückstände gelangen über verschiede Pfade in das Ökosystem und nicht alle davon sind biologisch abbaubar. Diese reichern sich in der Umwelt an und können in geringen Konzentrationen einen Einfluss auf Lebewesen haben.
Ziel dieser Arbeit ist die Anwendbarkeit des Verfahrens zur photolytischen Behandlung von Antiepileptika Levetiracetam (LEV) und Pregabalin (PB) durch den Einsatz von UV-Strahlung zu testen. Für die Abbauexperimente werden die Arzneimittel-Präparate Keppra® (LEV) und Lyrica® (PB) eingesetzt. Hierfür werden 100 mg/L des Wirkstoffes in Millipore-Wasser gelöst und zwei Stunden mit UV-Licht behandelt. Die Bestimmung der Primärelimination erfolgt mit der HPLC und der HPLC-MS.
Die Ergebnisse der Primärelimination zeigen eine kontinuierliche Abnahme der Konzentration der beiden Wirkstoffe ab 8 min mittels UV-Behandlung. Dabei werden Primärelimination von 59,8 ± 7,9 % (LEV) und von 60,3 ± 15,3 % (PB) erreicht. Beide Wirkstoffe werden kaum mineralisiert. Dies zeigt die Non Purgeable Organic Carbon (NPOC)-Bestimmung von 4,6 ± 1,9 % (LEV) und die Dissolved Organic Carbon (DOC)-Bestimmung von 5,4 ± 2,1 % (PB). Die anschließende kinetische Untersuchung der Abbauexperimente mit der HPLC-MS beweist die Bildung von Transformationsprodukten (TP). In beiden Experimenten entstehen instabile TP, die sich im Verlauf der Photolyse abbauen. Bei LEV entstehen 35 TP, von diesen sind 51,4 % polarer als die Muttersubstanz. Mit der HPLC-MS können Addukte mit Natrium nachgewiesen werden. Bei der Behandlung von LEV mit UV-Licht entstehen 12 TP, die für die toxikologische Untersuchung relevant sind.
Bei PB entstehen 16 potenzielle TP, von denen 43,8 % polarer als die Muttersubstanz sind. Bei PB kann im Vergleich zu LEV eine deutliche Zunahme des Molekülgewichtes nachgewiesen werden. Bei den kinetischen Untersuchungen können TP der Begleitsubstanz Lactose-Monohydrat zugeordnet werden. Für die Bewertung der Mineralisierung von PB wird ein unabhängiges Abbauexperiment mit Lactose-Monohydrat durchgeführt. Die voneinander unabhängig durchgeführten Abbauversuche belegen ein stabiles Eliminationsverfahren des Wirkstoffes LEV und PB. Für eine abschließende Beurteilung werden toxikologische Untersuchungen der TP gefordert sowie weitere Studien unter realen Umweltbedingungen empfohlen. Darüber hinaus muss eine Strukturaufklärung der TP erfolgen.

Residues of pharmaceuticals are transported via different ways into the ecosystem and not every drug is biodegradable. Those non-biodegradable drugs can be accumulated in the environment and may have an influence on living beings in low concentrations. The aim of this thesis is to test the applicability of the procedure of photolytic degradation to the anti-epileptic drugs Levetiracetam (LEV) and Pregabalin (PB) by UV radiation. For these degradation experiments the medicinal product Keppra® (LEV) and Lyrica® (PB) are used. For this purpose 100 mg/L of the active substance are solved in Millipore water. The solution will be treated two hours with UV light. The primary elimination is determined by means of the HPLC and the HPLC MS.
The results of the primary elimination show a continuous decrease of the concentration of both active substances starting after 8 min UV radiation. A primary elimination of 59.8 ± 7.9% (LEV) and from 60.3 ± 15.3% (PB) could be measured. Both active substances hardly mineralize. This is demonstrated by the Non Purgeable Organic Carbon (NPOC) determination of 4.6 ± 1.9% (LEV) and the Dissolved Organic Carbon (DOC) determination of 5.4 ± 2.1% (PB). The following kinetic examination of the experiments with the HPLC MS shows the formation of transformation products (TP). In both experiments unstable TP occurred which are decreased by photolysis. In case of LEV 35 TP are arising, thereof 51.4% are more polar than the mother substance. Using HPLC MS sodium adduct can be proven. Furthermore 12 TP are forming due to UV degradation of LEV which are relevant for the toxicological investigation.
In case of PB 16 potential TP are developing, thereof 43.8% are more polar than the mother substance. A comparison of PB to LEV shows a considerably increase of the molecule weight of PB. The kinetic examinations of the experiments show the formation of TP relating to the substance Lactose-mono hydrate. For the assessment of the mineralization of PB is carried out with an independent degradation experiment with lactose-mono hydrate.
The independently performed reduction experiments show a stable elimination process of the active substances LEV and PB. For a final assessment toxicological studies of the TP and testing under real environmental conditions are recommended. Additionally a structural clarification of the TP has to be done.
